Zimmerman, Leonard (U.S. Army Biological Laboratories, Fort Detrick, Frederick, Md.). Survival of Serratia marcescens after freeze-drying or aerosolization at unfavorable humidity. I. Effects of sugars. J. Bacteriol. 84:1297-1302. 1962.-Suspensions of Serratia marcescens were subjected to freeze-drying or to aerosolization at unfavorable humidity levels. The survival of the cells during one or the other of these treatments was markedly improved in the presence of common sugars, but no one sugar stabilized the cells against both stresses. The protective effects of the sugars were correlated with their penetrability into cells; minimally penetrable sugars stabilized cells against aerosolization, and freely penetrable sugars stabilized cells during freeze-drying. These results were attributed to the modifications of intracellular water content induced by the presence of the sugars in the cell suspensions.

Survival rate of microbes after freeze-drying and long-term storage.

The survival rates of 10 species of microorganisms were investigated after freeze-drying and preserving in a vacuum at 5 degrees C. The survival rates varied with species. The survival rates immediately after freeze-drying were different among yeast, gram-positive bacteria, and gram-negative bacteria, and the change in the 10-year survival rate was species-specific. The survival rate of yeast, ...

Relative humidity and the killing of bacteria. I. Observations on Escherichia coli and Micrococcus lysodeikticus.

Some observations on the ability of washed Escherichia coli and Mficrococcus lysodeikticus to form colonies when plated after having been freeze-dried and exposed to water vapor in vacuo will be presented in this paper. Serratia marcescens treated in this manner is rapidly killed within a rather narrowly limited range of relative humidities around 90 per cent (Monk and McCaffrey, 1957). It was ...

Preservation of microorganisms by freeze-drying. I. Cell supernatant, Naylor-Smith solution, and salts of various acids as stabilizers for Serratia marcescens.

Inasmuch as all living things usually require a high content of water either internally or externally, or both, it is remarkable that microorganisms, especially spore forms, can sometimes withstand an extreme desiccating environment. Any success achieved by chemical or physical means in modifying viability, that is in hastening or preventing death of the desiccated cell, has implications in man...
